We went to eat at the local Denny's (a 24 hour restaurant) a couple weeks ago, but they were closed. A sign on the door apologized for being closed until their water supply problem was fixed.
So yes, sometimes they do close.
But, normally they are open 24 hours a day, 7 days a week.

Some may close on certain holidays like Thanksgiving or Christmas, or may only be open for normal breakfast hours (until 10 or 11 AM local time) during those holidays, and then close for the remainder of the holiday so the employees can go home and spend the holiday with their families.

If the owners had a death in the family -- if it's a small family business, probably. If the place got robbed or was a crime scene, I'd think they'd close.
